Landover station opened on November 20, 1977, serving central Prince George's County in the heart of the Landover area. The station provides commuter rail access for a large suburban population along the US Route 50 corridor, one of the main east-west arteries of Prince George's County.

The station is primarily a commuter hub, with WMATA operating surface parking lots that allow riders from the broader Landover and Seat Pleasant area to park and take the Orange Line to downtown DC. Multiple Metrobus and Prince George's County TheBus routes also converge at the station, extending its effective service area well beyond walking distance.

The former Landover Mall site — one of the Washington area's largest shopping centers in its heyday — is located near the station.
